AB0011 EXPRESSION PROFILE AND POTENTIAL FUNCTION OF CIRCRNAS IN PERIPHERAL BLOOD MONONUCLEAR CELLS FROM PATIENTS WITH PRIMARY GOUT

Background: Autophagy is a phenomenon of “self-phagocytosis” in eukaryotic cells, which maintains cell homeostasis by transporting intracellular materials to lysosomes for degradation and recycling. In recent years, studies have shown that autophagy may be involved the pathogenesis rheumatoid arthritis(RA)[1], but its specific mechanism still unclear. Objectives: The expression levels autophagy-related genes(ATG) unc-51-like kinase 1(ULK1), ATG13, ATG17, microtubule associated protein 1 light chain 3 (LC3), P62 peripheral blood mononuclear cells (PBMC) patients with RA were detected, their role clinical significance explored. Methods: Real-time fluorescent quantitative PCR was performed detect ULK1, LC3, PBMCs 50 patients, healthy controls (HC), 25 moderate severe before after treatment. Then, t test, χ2 Mann-Whitney U Pearson test used statistical analysis. Results: 1.The hsCRP, white cell(WBC), neutrophils(GR), platelet(PLT) plateletcrit(PCT) group higher than those HC (P <0.05). Lymphocytes (LY), red cell(RBC), hemoglobin(HGB), hematocrit(HCT), mean corpuscular hemoglobin(MCH), volume(MCV) hemoglobin concentration(MCHC) lower 2.The expressions LC3 group, while group(P<0.05) (Figure 1). correlation analysis suggested ATG17 positively correlated tender joint count (TJC), swollen (SJC), health assessment questionnaire (HAQ) (P<0.05); ULK1 HAQ negatively (P<0.05).3. Compared treatment TNFi, HAQ, DAS-28, ESR, WBC, GR, PLT PCT significantly reduced RBC, HCT, MCV MCH increased treatment,(P<0.05); other related laboratory indicators not different TNFi (P>0.05).
